What 3 are the best to maintain in a standard group. I play the tank i get that there should be a mage rouge and healer who are the best characters to fill these rolls and if im wrong what set up should i keep?

Anders is your only healer after you lose your sister. Fenris would also be good for you to bring as your damage dealer, he works as a back up tank too. Varric is the best rogue, his archer tree is powerful and he's just fun to have around. But, to get the most out of the game you really shouldn't ignore anyone. The ones I listed are going to be the most useful to you, but you can play with almost any party setup.
Isabella and Merril are the only female love interests. You can flirt with Aveline, but it goes nowhere.
